Signal recognition particle 9 kDa protein (SRP9) is a small, conserved protein that functions as a core structural subunit of the signal recognition particle (SRP) complex, a ribonucleoprotein complex responsible for the cotranslational targeting and translocation of secretory and membrane proteins to the endoplasmic reticulum in eukaryotic cells. SRP9 forms a heterodimer with SRP14, binding to the Alu domain of SRP RNA, and is essential for the elongation arrest function of the SRP, helping to pause translation until the ribosome-mRNA-polypeptide complex can be properly engaged with the ER membrane translocon. Genetic studies have associated variation in SRP9 with susceptibility to motor neuritis, although its direct involvement in disease remains unclear. SRP9 plays an indirect but essential role in protein targeting fidelity and efficiency, with no evidence for direct targeting by therapeutic agents.
