Overview

Signal recognition particle receptor subunit beta (SRPRB) is a transmembrane GTPase and subunit of the ER membrane signal recognition particle (SRP) receptor complex. It anchors the alpha subunit of the SRP receptor to the endoplasmic reticulum membrane and is essential for the cotranslational targeting of secretory and membrane proteins. SRPRB coordinates the correct targeting, translocation, and N-glycosylation of nascent proteins at the ER. It plays a regulatory role in the assembly of the translocation supercomplex crucial for N-glycosylation, and its expression is involved in cell stress responses and the regulation of apoptosis by modulating NF-κB signaling. SRPRB expression and function have been implicated as a biomarker and potential therapeutic target in cancers such as pancreatic ductal adenocarcinoma, and it is directly targeted by the drug 6-thioguanine through its GTP-binding site, disrupting normal protein glycosylation and translocation[1][3][4][5][6][8].
