Signal transducers and activators of transcription (STATs) are a family of cytoplasmic transcription factors that mediate cellular responses to cytokines, growth factors, and other extracellular signals. Upon activation, STAT proteins translocate to the nucleus where they regulate gene expression involved in cell proliferation, differentiation, survival, inflammation, immune function, and oncogenesis. The canonical activation pathway involves receptor binding, JAK activation, STAT phosphorylation, dimerization, and nuclear translocation to regulate target gene transcription.
