Signal transducer and activator of transcription 5B (STAT5B) is a transcription factor and intracellular signaling molecule encoded by the STAT5B gene. It belongs to the STAT protein family, which transduces signals from cytokine and growth factor receptors to the nucleus, where it activates transcription of target genes. STAT5B is especially important for immune cell development, regulation of cell growth and apoptosis, and growth hormone signaling. Activation occurs via receptor-associated kinases, especially Janus kinases, which phosphorylate STAT5B, enabling dimer formation and nuclear translocation. STAT5B is implicated in several diseases, including cancers (notably leukemias), growth hormone insensitivity syndromes, and immune disorders. Therapeutically, the Jak-STAT pathway—including STAT5B—is targeted primarily by Janus kinase inhibitors for autoimmune and malignant conditions. Aberrant STAT5B activity can contribute to tumorigenesis, immune dysregulation, and allergic inflammation[1][2][4][6].
