Signal transducer and activator of transcription 5B (STAT5B) mRNA is the messenger RNA transcript that encodes the STAT5B protein, a key member of the STAT family of transcription factors. This mRNA is essential for the production of STAT5B, which mediates cellular responses to various cytokines and growth factors, including interleukin-2 (IL-2) and growth hormone (GH) (Source: UniProt P51692). Upon translation, the STAT5B protein is activated by Janus kinases (JAKs), leading to its phosphorylation, dimerization, and translocation to the nucleus where it regulates genes critical for cell survival and proliferation (Source: PubMed 28655779). Dysregulation or overexpression of STAT5B mRNA is a hallmark of several aggressive cancers, particularly T-cell prolymphocytic leukemia (T-PLL) and chronic myeloid leukemia (CML), where it promotes uncontrolled cell growth (Source: PubMed 31434683). Targeting the mRNA transcript directly using antisense oligonucleotides (ASOs) or RNA interference (RNAi) represents a promising therapeutic strategy to silence STAT5B expression, especially given the difficulty of targeting the protein's SH2 domain with small molecules (Source: PubMed 25605871). These RNA-based approaches aim to achieve high specificity and potency in reducing oncogenic STAT5B levels in patients with refractory malignancies (Source: NCBI Gene 6777).
