The STAT protein family consists of intracellular transcription factors that mediate cellular immunity, proliferation, apoptosis, and differentiation. They are activated by JAKs upon cytokine/growth factor stimulation, translocate to the nucleus, and regulate gene expression. There are seven mammalian STAT proteins (STAT1, 2, 3, 4, 5A, 5B, 6). Persistent activation is linked to cancer, while some STATs act as tumor suppressors. They interact with JAKs and other transcription factors like p53/NF-kB.
