Overview

Signal transducing adapter molecule 1 (STAM1) is an adaptor protein encoded by the *STAM* gene in humans[3][4]. It contains an SH3 domain and an immunoreceptor tyrosine-based activation motif (ITAM)[3]. STAM1 is rapidly phosphorylated in response to cytokine stimulation and mediates downstream signaling of cytokine receptors, associating with kinases like JAK2 and JAK3 via its ITAM region[3]. It interacts with HGS/HRS to form the ESCRT-0 complex, facilitating the recognition and sorting of ubiquitinated membrane proteins in the endosomal pathway for lysosomal degradation[1][4]. Additionally, STAM1 participates in ER to Golgi trafficking by interacting with the coat protein II complex[4]. Multiple alternatively spliced variants of STAM1 exist[4]. The protein is involved in pathways such as EGFR and Rho GTPase signaling, and interacts with several key partners involved in protein trafficking and receptor signaling, including HGS, MAP3K1, STAMBP, and TIMM8A[3][4]. Diseases associated with STAM1 include infectious diseases (such as Dengue virus infection) and neurodegenerative disorders[4]. There are no currently known drugs targeting STAM1 directly and no established use as a biomarker.
