Overview

Signal transducing adaptor molecule 2 (STAM2) is a multifunctional endosomal adaptor protein closely related to STAM1, involved in the downstream signaling of cytokine receptors through its SH3 domain and immunoreceptor tyrosine-based activation motif (ITAM)[1][3][5]. STAM2 acts downstream of JAK kinases, is phosphorylated in response to cytokine stimulation, and participates in the formation of the ESCRT-0 complex, essential for sorting ubiquitinated receptors into the multivesicular body pathway for lysosomal degradation[1][3][4][5]. It plays key roles in receptor down-regulation, regulation of cell signaling, and controlling cell proliferation. Deregulation or overexpression of STAM2 has been associated with tumorigenesis, particularly in gastric cancer where it promotes proliferation and metastasis through modulation of the JAK2/STAT3 pathway[3][5]. STAM2 may also have implications in neurodegenerative diseases such as frontotemporal dementia or ALS, and is expressed in various tissues including the immune system[6]. There are no known direct drug interactions or therapeutic agents targeting STAM2 as of now, but its pathway modulation capacity makes it a potential disease biomarker and a candidate for future therapeutic targeting in cancer and disorders of cellular signaling[3][5].
