Overview

Signaling lymphocytic activation molecule family member 3 (SLAMF3), also known as CD229 or Ly9, is a type I transmembrane glycoprotein belonging to the immunoglobulin superfamily and the SLAM family of receptors. Predominantly expressed on T cells, B cells, and NK cells, it features a distinctive extracellular domain with four immunoglobulin-like domains and a cytoplasmic tail with two immunoreceptor tyrosine-based switch motifs (ITSMs), which are essential for intracellular signaling. SLAMF3 functions as a self-ligand receptor modulating innate and adaptive immune responses through homophilic interactions, thereby playing important roles in immune activation, suppression, and regulation. It is implicated in a range of pathological conditions, including autoimmunity, cancer, and infections, and is being explored as a potential therapeutic target for immunomodulatory treatments.
