Overview

Signaling lymphocytic activation molecule family member 9 (SLAMF9) is a cell surface receptor belonging to the SLAM/CD2 family and the immunoglobulin superfamily[1][2][3][4]. It is expressed mainly on plasmacytoid dendritic cells (pDCs) and some macrophage populations, including tumor-associated macrophages[3][1]. Unlike other SLAM family members, SLAMF9 has a distinct short, lysine- and arginine-rich cytoplasmic tail. SLAMF9 regulates pDC differentiation, homeostasis, and activity, including production of type I interferons and TNF-α, and has significant roles in immune cell development and inflammatory responses[1][2][4]. Deficiency or dysfunction of SLAMF9 alters the maturation and abundance of pDCs, impacting both steady-state immune function and immune responses during inflammation or autoimmunity[1][2]. Targeting SLAMF9 is under consideration for modulating pDC-driven disease states, such as autoimmune conditions[4].
