The OT-I T-cell receptor (TCR) is a transgenic immune receptor that specifically recognizes the SIINFEKL peptide (residues 257–264 of ovalbumin) presented by the murine MHC class I molecule H-2Kb (Hogquist et al., 1994, Cell). It is composed of Vα2 and Vβ5.2 chains and serves as a foundational model in immunology for studying CD8+ T-cell development, activation, and memory (UniProt, 2024). In the context of drug discovery, the OT-I system is widely used to validate TCR-engineered T-cell (TCR-T) therapies and to assess the potency of antigen-presenting platforms like dendritic cell vaccines (PubMed, 2021). The receptor's high affinity for the H-2Kb-SIINFEKL complex allows for precise monitoring of T-cell responses, including cytokine production and cytotoxic activity against target cells (Nature Reviews Immunology, 2023). While primarily a murine research tool, it provides a critical framework for developing human TCR-based immunotherapies for cancer and infectious diseases (StatPearls, 2024).
The OT-I TCR binds specifically to the SIINFEKL peptide-H-2Kb MHC complex, initiating a signaling cascade through the CD3 complex and ZAP-70, which leads to T-cell proliferation, secretion of pro-inflammatory cytokines like IFN-gamma and TNF-alpha, and the release of perforin and granzymes to induce apoptosis in target cells (PubMed, 2021; Nature Reviews Immunology, 2023).
