Overview

The Silencing Mediator for Retinoid and Thyroid hormone receptor (SMRT) is a nuclear corepressor protein that plays a central role in the regulation of gene expression by nuclear receptors, including those for retinoids and thyroid hormones. SMRT functions primarily by repressing transcription in the absence of ligand binding to these receptors. It modulates adipocyte function by influencing adipogenesis, insulin sensitivity, leptin expression, lipolysis regulation via glucocorticoid signaling, and overall fat mass expansion. Altered expression levels have been observed in breast cancer tissues, and dysfunctional SMRT may contribute to obesity-related pathologies or type 2 diabetes risk profiles.
