SIM1 antisense RNA 1 (SIM1-AS1)

Target
SIM1-AS1
Molecular classification
Long non-coding RNA (lncRNA), Antisense RNA (asRNA)

Overview

SIM1 antisense RNA 1 (SIM1-AS1) is a long non-coding RNA transcribed antisense to the SIM1 gene locus. Like other antisense RNAs, it is thought to regulate gene expression primarily at the transcriptional or post-transcriptional level, including potential cis-acting effects on the nearby SIM1 gene. The molecular mechanisms may involve alteration of chromatin state, interference with transcription, or modulation of mRNA stability or translation. While antisense lncRNAs are increasingly recognized as critical regulators in diverse biological processes and diseases, SIM1-AS1 itself currently has no well-established role as a therapeutic target or disease biomarker.

Mechanism of action

Not applicable for SIM1-AS1 itself. Broadly, drugs targeting asRNAs (generally synthetic antisense oligonucleotides) act through: mRNA degradation (RNase H mediated), Steric hindrance of translation, or Modulation of splicing (general for antisense RNA targets, not specific for SIM1-AS1).

03

Biological functions

Regulation of gene expression (typically in cis, affecting expression of the SIM1 gene or nearby loci)Epigenetic regulation (can modulate chromatin state at the locus)RNA-mediated interference (by formation of asRNA-mRNA duplexes leading to translation inhibition or mRNA degradation, general asRNA property)
04

Disease associations

No specific diseases have been directly attributed to SIM1-AS1 itself in available databases as of the current literature. However, dysregulation of antisense lncRNAs can be implicated in cancer, developmental disease, and epigenetic disorders in general
05

Safety considerations

None specific to SIM1-AS1. In general, challenges for targeting asRNAs include off-target effects, difficulty in specific delivery, and unpredictable epigenetic consequences
