Overview

The Simian immunodeficiency virus (SIV) Gag polyprotein is a critical structural component of the SIV virion, serving as the primary driver of viral assembly and budding from infected cells (UniProt, P03354). During the viral life cycle, Gag is synthesized as a precursor (Pr55) that orchestrates the recruitment of viral genomic RNA and other components to the plasma membrane (PubMed, 25653451). Upon budding, the viral protease cleaves Gag into several functional subunits, including the matrix (MA), capsid (CA), nucleocapsid (NC), and p6 proteins, a process essential for viral maturation and infectivity (NCBI, V03332). Because of its high conservation and essential role in the viral life cycle, SIV Gag is a major target for the development of antiretroviral therapies, such as maturation inhibitors, and is a primary antigen used in experimental T-cell-based vaccines (PubMed, 23934130). In research settings, SIV Gag serves as a crucial model for understanding HIV-1 pathogenesis and testing novel therapeutic strategies for AIDS (StatPearls, NBK534860).
