Overview

Single-pass membrane and coiled-coil domain-containing protein 3 (SMCO3) is a human protein encoded by the SMCO3 gene, located on chromosome 12p12.3[1][5][7][10][13][15]. It consists of 225 amino acids, features a single transmembrane region, two coiled-coil regions, and contains the uncharacterized DUF4344 domain[1][15]. SMCO3 is predicted to localize primarily to the endoplasmic reticulum membrane, as indicated by its KKXX-like C-terminal motif[1]. It is expressed at low levels in several tissues, with highest expression observed in kidney, liver, and spleen, and increased expression in certain cancers such as chondrosarcoma and clear-cell renal cell carcinoma[1][7]. Functional characterization of SMCO3 is currently lacking, with no known biochemical or physiological role, and it is not implicated in any disease or recognized as a drug target[1][10][15]. Protein-protein interaction assays suggest association with proteins implicated in cancer and cell cycle regulation, but their biological significance is unclear[1]. No interacting drugs, known mechanisms of action, biomarker roles, or documented safety concerns are reported[1][7][10].
