Overview

Single-pass membrane and coiled-coil domain-containing protein 4 (SMCO4) is a human protein encoded by the SMCO4 gene (also known as C11orf75 or FN5), located on chromosome 11q13.3–q23.3[1][2][9][10]. SMCO4 is predicted to be a single-pass transmembrane protein with a coiled-coil domain and is present in the outer membrane of mitochondria[1][10]. Its structural features suggest it consists mainly of alpha-helices, with no known beta-turns or repeat elements[1]. The protein may be involved in processes related to protein–protein interaction, inferred from the presence of coiled-coil domains[6], and has predicted—though not experimentally confirmed—interactions with proteins such as BNIP3L, SYNE4, and CCDC155[1]. Expression is observed in numerous tissues with the highest levels in the salivary glands[1][8]. There is currently little direct evidence linking SMCO4 to therapeutic targeting, known drug interactions, biomarker status, or clear mechanisms relevant to human disease, and its molecular function remains largely uncharacterized[1][4][10].
