Overview

Single-stranded DNA-binding protein 2 (SSBP2) is a highly conserved protein essential for maintaining genomic integrity and regulating gene expression (UniProt P81877). It serves as a core component of the Sensor of Single-Strand DNA (SOSS) complex, which identifies and binds to single-stranded DNA at sites of damage to initiate the DNA damage response and facilitate telomere maintenance (PubMed: 19682734). Beyond its role in DNA repair, SSBP2 acts as a transcriptional cofactor by stabilizing the LIM domain-binding protein 1 (LDB1), thereby supporting the assembly of transcriptional complexes required for hematopoietic stem cell maintenance and differentiation (PubMed: 20473325). In clinical oncology, SSBP2 is primarily characterized as a tumor suppressor, with its loss or downregulation linked to the progression of various cancers, including acute myeloid leukemia, prostate cancer, and breast cancer (GeneCards). Additionally, SSBP2 is involved in oncogenic gene fusions, such as SSBP2-JAK2 and SSBP2-CSF1R, which are critical drivers in certain subtypes of acute lymphoblastic leukemia and are therapeutically targeted by kinase inhibitors like ruxolitinib (PubMed: 26912468). While direct small-molecule modulators of SSBP2 are not currently in clinical use, the protein is a significant biomarker for disease prognosis and a potential target for disrupting aberrant transcriptional complexes in cancer (PubMed: 33847955).
