Overview

Sirtuins are a highly conserved family of proteins found across all domains of life. They are NAD+-dependent deacetylases and ADP-ribosyltransferases that regulate a variety of cellular processes, including gene expression, DNA repair, metabolism, stress responses, and aging. In mammals, there are seven sirtuins (SIRT1–SIRT7), each with distinct cellular localizations and biological functions. Sirtuins are considered promising pharmacological targets for treating age-associated diseases or extending healthy lifespan.

Mechanism of action

Sirtuins function primarily as NAD+-dependent deacetylases and ADP-ribosyltransferases. They remove acetyl groups from lysine residues on histones and various non-histone proteins, thereby regulating gene expression, DNA repair, metabolism, stress responses, apoptosis, and aging processes.
