Sirtuin 6 (SIRT6) is a nuclear protein that functions as a NAD+-dependent deacetylase, mono-ADP-ribosyltransferase, and defatty-acylase. It plays crucial roles in DNA repair, aging, metabolic regulation, and inflammation. SIRT6 deacetylates histones H3K9ac and H3K56ac, influencing chromatin structure and gene expression. It is essential for base excision repair and double-strand break repair. Overexpression can extend lifespan, while deficiency leads to premature aging phenotypes and increased cancer risk. SIRT6 regulates glucose and lipid metabolism and modulates inflammatory responses by controlling cytokine secretion. Targeted modulation of SIRT6 activity is being explored for the treatment and prevention of age-related diseases, cancer, cardiovascular diseases, and metabolic disorders.
