Sjögren syndrome nuclear autoantigen 1 (SSNA1), also known as NA14, is a small (~14 kDa) coiled-coil microtubule-associated protein that plays critical roles in the regulation of microtubule dynamics. SSNA1 stabilizes microtubules by suppressing dynamic instability parameters: it slows the rates of microtubule growth, shrinkage, and catastrophe, while promoting rescue events. SSNA1 binds to both microtubule ends and the lattice, forms fibrillar assemblies, and is highly enriched at sites of microtubule lattice damage—both naturally occurring and those induced by microtubule-severing enzymes such as spastin. Functionally, SSNA1 is involved in processes such as cilia formation (localizing to basal bodies and axonemes), cell division (enrichment at spindle poles and midbodies), and neural development (promoting axon elongation and branching). Its role as a target antigen in Sjögren’s syndrome accounts for its autoantigenic relevance, but it is not currently known to be a direct target of approved drugs or therapeutic interventions[1][2][3].
