SKI family transcriptional corepressor 2 (SKOR2) is a transcriptional corepressor and member of the SKI family, enabling SMAD binding and sequence-specific double-stranded DNA binding activities[1][2]. SKOR2 is principally expressed in the nervous system, particularly marking Purkinje cells in the cerebellum, where it regulates gene expression critical for cell-type identity and development[1][4]. The protein acts as a negative regulator of the TGF-β signaling pathway by repressing SMAD-dependent transcription, thus acting as a TGF-β antagonist in neuronal tissues[1][5][6]. Although not recognized as a therapeutic target, alterations in SKOR2 expression or methylation have been associated with abnormal cerebellar development in animal models, and methylation changes have been described in some cancers, but its role as a disease driver remains unclear[1][4][5]. Currently, there are no approved drugs that target SKOR2, nor is it used as a biomarker or direct therapeutic target in human medicine[1][2].
