Overview

Ski2-like RNA helicase (SKIC2) is a cytoplasmic RNA helicase belonging to the DExH-box family, serving as a core component of the SKI complex, which collaborates with the cytoplasmic exosome to facilitate turnover and quality control of mRNAs[1][2][3]. SKIC2 is highly conserved across eukaryotes and is implicated in the extraction and degradation of aberrant or excess mRNA via 3'-to-5' decay by unwinding RNA substrates and channeling them to the exosome[1][2]. In humans, loss-of-function variants in SKIC2 cause trichohepatoenteric syndrome, a rare disorder affecting multiple organs, presumably by impairing cellular RNA quality control pathways[3]. This protein may also have a role in cellular antiviral defense by targeting viral RNAs for degradation[2].
