Physical barrier function refers to the collective mechanical and biochemical protective mechanisms provided primarily by the skin and mucous membranes. This function relies on the structure and integrity of the skin, especially the outermost layer (stratum corneum), which is composed of densely packed corneocytes embedded in a lipid matrix of ceramides, fatty acids, and cholesterol. The physical barrier serves as the body's first line of defense, blocking the entry of pathogens, harmful chemicals, and mechanical insults, while also preventing excessive water loss and helping regulate body temperature. Impairment of physical barrier function can result in increased susceptibility to infections, inflammatory skin diseases (such as atopic dermatitis), and loss of essential moisture[1][3][4][7][9][10]. Note: "Physical barrier function" is *not* a molecule, receptor, or conventional drug target, but rather a broad physiological role performed by tissues comprising various molecular elements. It does not correspond to a single therapeutic target suitable for molecular drug design.
