Overview

Skin extracellular matrix (ECM) water content is a physiological parameter representing the hydration status of the dermal and epidermal compartments, rather than a single molecular target. It is primarily maintained by the hygroscopic properties of ECM components, particularly hyaluronic acid and other glycosaminoglycans, which sequester water to provide tissue turgor and elasticity (Source: Papakonstantinou et al., 2012). This hydration is critical for the skin's barrier function, thermoregulation, and the diffusion of signaling molecules and nutrients (Source: Verdier-Sévrain & Bonté, 2007). Clinically, reduced skin water content is a hallmark of aging and various dermatological conditions, including xerosis and atopic dermatitis, where it contributes to pruritus and inflammation. While pharmacological interventions do not target "water content" as a protein, they aim to modulate it through the application of humectants, occlusives, or agents that stimulate the synthesis of water-binding ECM molecules. Consequently, it serves as a vital efficacy endpoint in the development of topical and systemic dermatological treatments.
