Skin innate immune receptors refer to a broad class of pattern recognition receptors (PRRs) expressed by various cells in the skin, including keratinocytes, Langerhans cells, and dermal dendritic cells. These receptors, which include Toll-like receptors (TLRs), NOD-like receptors (NLRs), and C-type lectin receptors (CLRs), serve as the first line of defense by detecting pathogen-associated molecular patterns (PAMPs) and danger-associated molecular patterns (DAMPs). Upon activation, they trigger signaling cascades that lead to the production of pro-inflammatory cytokines, chemokines, and antimicrobial peptides. While essential for host defense against infections, dysregulation of these receptors is heavily implicated in the pathogenesis of chronic inflammatory skin conditions such as psoriasis and atopic dermatitis. Because this term encompasses a wide variety of distinct protein families with different structures and functions, it is considered a functional category rather than a specific therapeutic target.
