Overview

Skin melanocytes are specialized pigment-producing cells derived from neural crest cells that reside primarily in the basal layer of the epidermis and hair follicles, where they synthesize melanin within melanosomes through the process of melanogenesis.[1][2][3] These dendritic cells transfer mature melanosomes to surrounding keratinocytes, forming a protective cap over the nucleus that shields DNA from ultraviolet (UV) radiation damage, thereby determining skin and hair color and providing photoprotection.[1][4] Melanocytes also function as immune sentinels, exhibiting phagocytic activity, antigen presentation via MHC class II, and secretion of proinflammatory cytokines such as IL-1, IL-6, TNF-α, and TGF-β in response to pathogens or stressors.[1][2] Their development involves migration of melanoblasts from the neural crest, regulated by pathways like KIT/SCF, Wnt/β-catenin, and MITF transcription factor, which controls differentiation, survival, and pigmentation genes including TYR, TYRP1, and DCT.[2][3] Dysregulation of melanocyte function or number contributes to pigmentation disorders like vitiligo (loss of melanocytes) and melanoma (malignant proliferation), highlighting their role in skin homeostasis and disease.[3][5] While not a specific molecular therapeutic target like receptors or enzymes, melanocytes are indirectly modulated in dermatological therapies targeting pigmentation pathways.[1][3]
