Overview

The skin neurosensory system is an integrated network of sensory nerve fibers, specialized receptors, and skin cells that detect and respond to environmental stimuli such as pressure, temperature, and chemical irritants (Bousbiat et al., 2023, PMID: 37443639). It facilitates communication between the skin and the central nervous system through the release of neuropeptides and the activation of various ion channels, such as Transient Receptor Potential (TRP) channels (Misery et al., 2016, PMID: 27117193). Key components include A-delta and C-fiber sensory neurons, Merkel cells, and their interactions with keratinocytes and immune cells. Dysregulation of this system is a hallmark of conditions like chronic pruritus, sensitive skin, and atopic dermatitis, where neurogenic inflammation exacerbates tissue damage and discomfort (Steinhoff et al., 2014, PMID: 24506931). Because it encompasses a broad array of cell types and signaling pathways rather than a single protein, it is considered a physiological system rather than a discrete molecular drug target. Therapeutic interventions often focus on specific nodes within this system, such as TRPV1 or neuropeptide receptors, to modulate sensory output and inflammatory responses.
