Overview
The skin's physical barrier, primarily the stratum corneum, is the outermost layer of the epidermis. It consists of dead corneocytes embedded in a lipid matrix, forming a 'bricks and mortar' structure. This barrier protects against pathogens, toxins, UV radiation, and prevents excessive water loss. Key components include corneocytes, lipids (ceramides, cholesterol, fatty acids), filaggrin, and the acid mantle. Disruption of the skin barrier is implicated in various skin conditions like eczema, psoriasis, and contact dermatitis, leading to dryness, inflammation, and increased permeability.
