Skin proteins involved in hapten formation are endogenous proteins within the skin that undergo covalent modification by small reactive chemicals known as haptens. This process, called protein haptenation, is a critical molecular initiating event in the development of allergic contact dermatitis and other hypersensitivity reactions. Haptens are typically low-molecular-weight electrophilic chemicals that covalently bind to nucleophilic amino acid residues (such as lysine, cysteine, histidine, and tyrosine) on skin proteins. The resulting hapten-protein conjugates (neoantigens) can be recognized by the immune system as foreign, triggering an immune response and inflammation. This process is central to allergic contact dermatitis and other forms of drug-induced or chemical-induced hypersensitivity. Protein haptenation is the basis for in vitro assays that predict skin sensitization potential.
