Overview

Skin structural proteins constitute a diverse group of molecules, including keratins, collagens, and elastin, which are fundamental to the mechanical integrity and physiological function of the skin (StatPearls, 2023). Keratins form the primary cytoskeletal framework of the epidermis, while filaggrin is essential for aggregating these filaments to maintain the stratum corneum barrier (PubMed, PMID: 29411831). In the dermis, Type I and Type III collagens provide tensile strength, and elastin fibers allow the skin to return to its original shape after stretching (NIH, 2022). These proteins are critical in the pathology of various conditions; for instance, collagen degradation is a hallmark of skin aging, and mutations in filaggrin are strongly linked to atopic dermatitis (PubMed, PMID: 16550169). Pharmacological interventions often target these proteins indirectly; retinoids like tretinoin are well-documented to stimulate collagen synthesis and improve skin texture (PubMed, PMID: 17515510). Additionally, topical emollients and humectants are used to support the structural barrier formed by these proteins in xerotic and inflammatory skin diseases. Because this category encompasses multiple distinct protein families, it is considered a non-specific therapeutic target grouping rather than a single molecular entity.
