Overview

SKN-1 (Skinhead-1) is a transcription factor in the nematode Caenorhabditis elegans that serves as the functional and structural ortholog of the mammalian Nrf (Nuclear factor erythroid 2-related factor) family, specifically Nrf2. It is a master regulator of the organism's cytoprotective response, orchestrating the expression of antioxidant and Phase II detoxification genes in response to oxidative and xenobiotic stress (Blackwell et al., 2015). Beyond its role in stress resistance, SKN-1 is essential for early embryonic development, where it specifies the fate of mesendodermal cells, and it significantly influences lifespan and metabolic homeostasis (An & Blackwell, 2003). In the context of drug discovery, SKN-1 is frequently utilized as a model target to identify and validate small molecules, such as sulforaphane and dimethyl fumarate, that activate the conserved Nrf2 pathway to combat age-related decline (Robida-Stubbs et al., 2012). Therapeutic interest in the SKN-1/Nrf2 pathway stems from its potential to mitigate damage in neurodegenerative diseases, chronic inflammation, and metabolic disorders by enhancing cellular resilience. However, the pathway presents a "double-edged sword" in oncology, as its activation can protect healthy cells from carcinogens but may also provide a survival advantage to established cancer cells, leading to drug resistance (Blackwell et al., 2015). Consequently, while SKN-1 activators are sought for their anti-aging and neuroprotective properties, careful consideration of the cellular context and the risk of promoting tumorigenesis is required. Monitoring SKN-1 activity is typically achieved through the induction of downstream targets like GST-4, which serves as a reliable biomarker for pathway engagement (Onken & Driscoll, 2010).

Mechanism of action

Activation of cytoprotective gene expression through binding to antioxidant response elements (ARE) or SKN-1 binding sites (SBS) following nuclear translocation, often mediated by the inhibition of negative regulators like WDR-23 or Keap1 (Blackwell et al., 2015).
