Overview

SLAMF6 is a type I transmembrane protein belonging to the CD2 subfamily within the immunoglobulin superfamily. It functions as a self-ligand receptor mediating homotypic interactions between immune cells and acts as a co-receptor in T cell and NK cell activation and inhibition. Its expression is high on T lymphocytes, NK cells, B lymphocytes, and dendritic cells. SLAMF6 interacts with SH2 domain-containing proteins such as SAP/SH2D1A and phosphatases like SHP family members, modulating downstream signaling. It plays a role in tumor immunity, with its absence augmenting tumor killing by CD8+ T cells. It is being explored as a therapeutic target to enhance anti-cancer immunity, but potential safety concerns include immune-related adverse events and autoimmunity.
