Overview

SLAM family member 8 (SLAMF8) is a cell surface receptor, classified within the CD2/SLAM receptor family, and primarily expressed on macrophages, neutrophils, dendritic cells, and some lymphocyte populations. It contains immunoglobulin-like domains and mediates cell-cell signaling that modulates immune cell activation, differentiation, and inflammatory response. Functionally, SLAMF8 negatively regulates activation and microbicidal function in macrophages via suppression of NADPH oxidase activity, and impacts multiple immune processes, including B cell development and T cell costimulation. Recent research highlights SLAMF8 as a tumor-infiltrating immune biomarker and a potential novel immunotherapy target in cancer due to its role in shaping the tumor microenvironment, especially by influencing macrophage phenotype and immunosuppressive signaling. No direct endogenous ligand has been identified, and there are no approved drugs specifically targeting SLAMF8; however, some kinase inhibitors can indirectly impact its function in certain cancer context.
