SLC16A1 antisense RNA 1 (SLC16A1-AS1) is a long non-coding RNA transcribed antisense to the SLC16A1 gene (lactate transporter MCT1) and serves as a regulatory molecule involved in metabolic reprogramming of cancer cells. It forms complexes with transcription factors such as E2F1, facilitating upregulation of SLC16A1/MCT1 and genes like PPARA, thus promoting a metabolic phenotype favoring cancer cell invasiveness. SLC16A1-AS1 is regulated in a cancer-type-specific manner and acts by modulating microRNAs and gene stability, making it a promising biomarker and candidate therapeutic target in oncology. It does not encode a protein and is not a classical drug target (such as receptor or enzyme), but may be targeted by RNA therapeutics.
Gene silencing with antisense oligonucleotides or CRISPR-Cas9 disrupts SLC16A1-AS1, reducing cancer cell invasiveness. It also involves potential microRNA modulation, such as sponging miR-143-3p (renal cancer) and miR-515-5p (colorectal cancer).
