Overview

SLC25A25 antisense RNA 1 (SLC25A25-AS1) is a cytoplasmic long non-coding RNA that functions as an oncogenic regulator in several cancer types, including non-small cell lung cancer, gastric cancer, and colorectal cancer[2][4][5]. It modulates cell proliferation, apoptosis, migration, and invasion, primarily by acting as a competitive endogenous RNA that sequesters specific microRNAs such as miR-195-5p. Through this mechanism, SLC25A25-AS1 upregulates downstream targets including integrin α2 (ITGA2), and contributes to epithelial-mesenchymal transition and tumor progression. SLC25A25-AS1 expression levels are associated with adverse prognostic features in NSCLC and may be used as a biomarker for patient stratification. No drugs currently directly target SLC25A25-AS1, but it represents a promising experimental therapeutic target in RNA-based cancer treatments[2][5][4].

Mechanism of action

Antisense therapeutics or siRNA: silences SLC25A25-AS1, resulting in decreased cell proliferation, migration, and invasion, and increased apoptosis in cancer cells. lnRNA functions as a ceRNA: SLC25A25-AS1 sponges miR-195-5p, thereby increasing ITGA2 expression, which promotes tumorigenic activity.
