Overview

SLC2A1 antisense RNA 1 (SLC2A1-DT) is a long non-coding RNA transcript located divergently to the SLC2A1 gene, which encodes the glucose transporter GLUT1. SLC2A1-DT is highly expressed in hepatocellular carcinoma, promotes tumor cell glycolysis and proliferation, and is mechanistically involved in stabilizing β-catenin by modulating its interaction with YWHAZ, thus enhancing β-catenin/c-Myc-dependent transcription of glycolytic genes (including SLC2A1, LDHA, and HK2). Its stability is further increased via m6A RNA methylation by METTL3 in a c-Myc-dependent feed-forward loop. SLC2A1-DT is emerging as a candidate therapeutic target and disease biomarker in glycolysis-driven cancers, especially HCC.
