Overview

SLC44A3 antisense RNA 1 (SLC44A3-AS1), also known as LINC01057, is a long non-coding RNA (lncRNA) transcribed antisense to the SLC44A3 gene. It does not code for a protein but regulates gene expression through interactions with proteins and other regulatory molecules. SLC44A3-AS1 is notably overexpressed in glioblastoma, where it activates NF-κB signaling by interacting with IKKα, maintains chromatin accessibility at NF-κB responsive promoters, and promotes mesenchymal transition—an aggressive and therapy-resistant cellular phenotype. Its high expression correlates with poor prognosis in mesenchymal-subtype glioblastoma, and knockdown studies indicate it has roles in cell proliferation, invasion, and resistance to radiation therapy. Current research supports consideration of SLC44A3-AS1 as a potential therapeutic target and biomarker in cancer, particularly high-grade gliomas[1][5].

Mechanism of action

As a non-coding RNA, therapeutic interventions may involve antisense oligonucleotides or RNA interference, but no specific small molecules or drugs directly acting on SLC44A3-AS1 are described in current literature[2].
