Overview

SLIT and NTRK-like family member 3 (SLITRK3) is a single-pass transmembrane synaptic cell adhesion molecule highly enriched at inhibitory synapses, where it promotes the differentiation and maintenance of inhibitory (GABAergic) synapses in the central nervous system. It interacts extracellularly with presynaptic receptor protein tyrosine phosphatase δ (PTPδ) and postsynaptically with neuroligin 2 (NL2), as well as intracellularly with gephyrin to stabilize synaptic architecture. Variants in SLITRK3 are implicated in neurodevelopmental disorders, especially those with epileptic features, and its expression is investigated as a prognostic marker in some cancers. SLITRK3 is not a classical neurotransmitter receptor or ion channel, but a molecular organizer guiding synapse formation and maturation.
