SLIT and NTRK-like protein 2 (SLITRK2) is a single-pass transmembrane protein predominantly expressed in postsynaptic neurons, functioning as a synaptic cell-adhesion molecule. It promotes neurite outgrowth, regulates the development and maintenance of excitatory synapses, and is involved in synaptogenesis. The extracellular region of SLITRK2 comprises two N-terminal leucine-rich repeat (LRR) domains, structurally similar to those in SLIT proteins, which mediate binding with receptor protein tyrosine phosphatases (notably PTPδ). Genetic variants in SLITRK2 are implicated in X-linked neurodevelopmental disorders, with pathogenic mutations disrupting synaptic signaling and neuronal circuit integrity. No current drugs are described that target SLITRK2 directly, and due to its essential neurological functions, therapeutic manipulation may carry risks of cognitive or behavioral side effects.
