Slit guidance ligand 1 (SLIT1) is a secreted extracellular matrix protein that serves as a molecular guidance cue, primarily for axons and neural precursor cells in the central nervous system[1][2][3][4]. It binds to Roundabout (ROBO) family receptors, mediating repulsive signaling that directs axonal growth cones away from the midline during neural development and restricts inappropriate axon crossing. SLIT1 is also involved in regulating SVZ-derived precursor cell migration in adult neurogenesis and CNS repair, especially following injuries such as demyelination[2]. Loss of SLIT1 results in increased ectopic dispersal and recruitment of progenitor cells to lesion sites, but does not directly affect precursor proliferation or survival. SLIT1 is widely expressed in brain tissues, with a crucial role in neuron projection morphogenesis, chemorepulsion, and neurodevelopmental patterning[1][2][4]. Currently, SLIT1 has no approved interacting drugs, but its role in CNS repair makes it a potential therapeutic target under investigation for neurodegenerative and demyelinating diseases[2].
