Slit guidance ligand 3 (SLIT3) is a secreted glycoprotein that belongs to the Slit family, characterized by four leucine-rich repeat (LRR) domains, several epidermal growth factor (EGF)-like domains, and a C-terminal cystine knot[1][8]. SLIT3 primarily functions as a ligand for Robo family receptors (notably Robo1 and Robo4). It serves crucial roles in axon guidance in the nervous system, where it acts as a midline repellent, but is also essential for non-neuronal processes such as promoting angiogenesis, bone remodeling, and sympathetic innervation of brown adipose tissue[4][2][6]. SLIT3 is highly conserved and predominantly expressed in the thyroid, endothelium, brown adipose tissue, and bone. Genetic variation and altered expression of SLIT3 are associated with osteoporosis, vascular disorders, neurodevelopmental diseases, and metabolic dysfunctions[10][4][5][6]. Its biological activities make it an emerging target in angiogenesis, neurodevelopment, and metabolic research, but no approved drugs currently target SLIT3 directly.
