SLIT2 intronic transcript 1 (SLIT2-IT1) is a long non-coding RNA (lncRNA) located within an intron of the SLIT2 gene. It does not encode a protein and instead exerts regulatory effects at the RNA level. SLIT2-IT1 expression is epigenetically regulated by SLIT2 promoter methylation: promoter hypermethylation represses both SLIT2-IT1 and the related microRNA miR-218[2]. In hematologic malignancies such as myelodysplastic syndrome, acute myeloid leukemia, and chronic myeloid leukemia, repressed SLIT2-IT1 expression is observed with promoter hypermethylation and may be linked to disease progression and poor clinical outcomes[2]. High SLIT2-IT1 expression may enhance cell proliferation, colony formation, mitotic activity, and resistance to apoptosis in the context of leukemogenesis[1]. However, SLIT2-IT1 remains poorly characterized and is not considered a classical drug target such as a receptor, enzyme, or transporter. Its main role is as a potential biomarker and a component of the non-coding transcriptome implicated in oncogenic regulation[1][2]. There are no known drugs or specific mechanisms of action identified for targeting SLIT2-IT1.
