Smad family member 8 (SMAD8), also known as SMAD9 or MADH6, is a receptor-regulated SMAD (R-SMAD) that mediates signaling by bone morphogenetic proteins (BMPs). Upon activation by BMP receptors, SMAD8 forms complexes with co-Smad4 and translocates to the nucleus to regulate target gene expression. It plays a critical role in cell growth, apoptosis, morphogenesis, development, and immune responses. The protein's activity is tightly regulated through phosphorylation and ubiquitination, ensuring precise control over BMP-mediated gene expression.
