Overview

Small Cajal body-specific RNA 21 (SCARNA21/ACA68) is a non-coding RNA molecule localized to Cajal bodies where it guides the conversion of specific uridine residues to pseudouridine in U12 snRNA, a minor spliceosome component[2]. It belongs to the H/ACA box snoRNA family and plays a crucial role in ensuring the correct maturation and modification of spliceosomal RNAs, which are essential for RNA splicing[2][5]. SCARNA21 is not considered a conventional therapeutic target. Although snoRNAs and scaRNAs have been implicated as molecular markers in cancer and other diseases, there are no reports of SCARNA21 itself being directly linked to a specific pathology or being targeted by drugs[6].

Biological functions

Guide pseudouridylation of uridine residues to pseudouridine in minor spliceosomal RNAs (especially U12 snRNA)Facilitate maturation and modification of spliceosomal RNAs in the Cajal bodySupport accurate and efficient splicing of pre-mRNA by modifying snRNAs

Disease associations

Other (there is emerging evidence that dysregulation of snoRNAs—including scaRNAs—may act as molecular markers in cancer and other diseases, but SCARNA21-specific roles are not well established)
