Small cysteine and glycine repeat-containing protein 5 (SCYGR5), also known as keratin-associated protein 28-5 (KRTAP28-5), is a cysteine-rich protein highly expressed in the hair cortex, where it forms part of the interfilamentous matrix embedding hair keratin intermediate filaments. These keratin-associated proteins are essential for generating a rigid and resilient hair shaft, primarily by cross-linking with hair keratins via disulfide bonds due to their cysteine content. SCYGR5 belongs to the larger family of keratin-associated proteins (KRTAPs), which are not receptors, enzymes, or therapeutic targets in current medical research. SCYGR5 does not have known roles in disease or as a drug target, and its primary biological function is structural in the differentiation of hair fibers. SCYGR5 is an 85-amino-acid protein, molecular weight ~8.2 kDa. It is located predominantly within the intermediate filament of hair cortex cells. No literature supports a direct role in disease, therapy, or biomarker applications. There is no evidence that SCYGR5 is a therapeutic target, receptor, enzyme, transporter, or member of any major signal transduction pathway. Its function is tightly restricted to hair fiber structure.
