Overview

Small G protein signaling modulator 2 (SGSM2) is a protein that belongs to the SGSM family, characterized by the presence of both RUN and TBC domains, and it functions as a GTPase-activating protein (GAP) for Rab32 and Rab33B, which are involved in regulating membrane trafficking[1][7]. SGSM2 modulates small G protein-mediated signaling pathways, with roles in both neuronal processes (including vesicle trafficking) and cancer biology[1][3][5]. It interacts with key regulators such as E-cadherin/β-catenin at the plasma membrane, influencing cytoskeletal dynamics, cell adhesion, and migration[3]. SGSM2 has been shown to act as a tumor suppressor in thyroid and breast cancer by activating RAP1 to inhibit RAS-mediated signaling, thereby reducing cell proliferation, invasion, and migration[5]. Additionally, gene expression analyses have identified SGSM2 as relevant in inflammatory and metabolic pathways, and as a hub gene in mood disorders such as subsyndromal symptomatic depression[1]. As of now, there are no known drugs directly targeting SGSM2, and no substantial clinical safety concerns have been reported, as it is not yet an established therapeutic target[1][5][7].
