Overview

Small G protein signaling modulator 3 (SGSM3) is a multi-domain adapter protein classified within the RUN and TBC1 domain-containing family, functioning primarily as a modulator of small G protein signaling pathways involving RAP and RAB subfamilies[1][2]. It interacts with connexin 43 (Cx43), a key gap junction protein, modulating its turnover and cellular localization, and thereby participating in the regulation of cell–cell communication, apoptosis, and cell differentiation, notably in cardiac and stem cell contexts[1]. Variants in SGSM3 have been associated with autism spectrum disorders and various cancers[1][2]. The protein is expressed in multiple tissues including the central nervous system, and while mechanistic and drug interaction data remain limited, it is regarded as a potentially important modulator of cell signaling and survival, with roles emerging in cardiac biology, neurodevelopment, and tumorigenesis[1][2].
