Small integral membrane protein 3 (SMIM3) is a single-pass transmembrane protein encoded by the SMIM3 gene located on human chromosome 5q33.1. It consists of 60 amino acids, with highest expression in the heart, ovary, and adrenal glands, and lowest in skeletal muscle and cerebellum[2]. Its precise physiological function is not completely characterized, but evidence suggests roles in cell channel regulation, neuronal differentiation, and cancer biology[2]. In acute myeloid leukemia cells, SMIM3 is overexpressed and predominantly localizes to the nucleus and intracellular vesicles[2]. Knockdown of SMIM3 inhibits proliferation, causes cell cycle arrest (G0/G1), and induces apoptosis via upregulation of p27 and downregulation of Cyclin D1 and CDK4[2]. Cellular and pathway analyses indicate association with ion channel complexes, Wnt and GPCR binding, and the PI3K-AKT signaling cascade[2]. Higher SMIM3 expression correlates with poor prognosis in certain cancers and with sensitivity to PI3K-AKT pathway inhibitors and some antineoplastic drugs[2]. SMIM3 is also suggested as a sensitive and specific biomarker for radiation exposure, though it is not currently established as a therapeutic target or direct drug receptor[2][4][6].
