Small integral membrane protein 42 (SMIM42) is a putative protein encoded by the *SMIM42* gene, predicted to contain a single 21-amino-acid transmembrane helix. It is composed of 70 amino acids in total and is classified as a small membrane protein based on structural modeling and sequence analysis. While there is transcriptional evidence (e.g., RNA-seq) supporting testis-specific expression, there is currently no proteomic or mass spectrometry evidence confirming that SMIM42 is indeed translated in vivo. Conservation of the open reading frame is seen in placental mammals, but several mammalian lineages have lost it. The function, biological significance, and therapeutic relevance of SMIM42 remain uncertain. It is included in some gene and protein annotation sets but has been excluded from the highest-confidence lists of functional small proteins due to ambiguous coding status and lack of functional data.
