Overview

Small nucleolar RNA host gene 6 (SNHG6) is a long non-coding RNA highly expressed in various cancers, where it acts primarily as a competing endogenous RNA (ceRNA) by "sponging" tumor-suppressor microRNAs, thus derepressing oncogenic targets and promoting proliferation, migration, invasion, angiogenesis, and resistance to apoptosis. SNHG6 also interacts with RNA-binding proteins, notably hnRNPA1, to influence alternative splicing of pyruvate kinase M (PKM), favoring PKM2 expression and supporting cancer cell aerobic glycolysis. Furthermore, SNHG6 modulates diverse signaling pathways, including TGF-β/Smad, PI3K/Akt/mTOR, JAK2/STAT3, and the Wnt/β-catenin pathway, contributing to tumor progression, metastatic potential, and drug resistance in multiple malignancies. Its expression and activity are being explored as prognostic biomarkers and potential therapeutic targets, but the complexity of its molecular interactions presents challenges for clinical translation.
